Introduction: Cross-infection, a significant concern in healthcare settings, is often associated with insufficient sterilization of medical tools and surfaces, where Staphylococcus aureus plays role in nosocomial infections. To prevent cross-infection, use of disinfectants before and after medical procedures is crucial. This study’s aimed to evaluate combined infusion of Betel Leaf and Lemongrass Stem as a potential disinfectant strategy to combat S. aureus. Materials and methods: A post-test control group design for a laboratory experiment, consisted of five treatment groups: betel leaf, lemongrass stem, a combination of green betel leaf and lemongrass stem infusion, positive control using sodium hypochlorite, and negative control using aquadest. Assessment of bacterial growth inhibition zones on Muller-Hinton agar involved measuring zone diameters. Statistical analysis included Kruskal-Wallis test (p < 0.05) followed by Mann-Whitney test. Results: Findings revealed beneficial alkaloids, saponins, tannins, and flavonoids in both lemongrass and green betel leaf, demonstrating antibacterial properties. The combination infusion exhibited an average increased inhibitory zone of 7.23 mm, showcasing enhanced antibacterial efficacy. Conclusion: The combined infusion of green betel leaf and lemongrass stem exhibited promising effectiveness as a disinfectant against S.aureus, The study highlights natural plant potential as alternative disinfection agents in healthcare settings to mitigate cross-infection risks.
